제품 설명
flashBAC™ Baculovirus Expression System은 baculovirus를 이용한 단백질 발현 시스템으로, baculovirus DNA와 control transfer plasmid로 구성되어 있다.
본 시스템은 Autographa californica multiple nucleopolyhedrovirus (AcMNPV)의 genome에서 필수 유전자 ORF1629를 결실시켜 곤충세포내에서 재조합되지 않은 virus가 복제되지 않으므로, plaque assay를 진행하지 않고도 재조합된 virus만을 얻을 수 있다.
또한 Chitinase gene (chiA)를 제거하여 분비형 단백질이나 막단백질을 매우 효율적으로 발현시킬 수 있다.
flashBAC™ ULTRA Baculovirus Expression System (Code MIR 6135, MIR 6140)는 필수적이지 않은 유전자를 제거함으로써 재조합 단백질의 발현양을 높인 업그레이드 시스템이다.
이 시스템은 AcMNPV polyhedrin 유전자 부위에 bacterial artificial chromosome (BAC)이 삽입되어 있다.

Description

flashBAC™

The original flashBAC™ expression vector. Protein expression and secretion is improved by deletion of the chitinase gene from the AcMNPV genome. Optimal for expression of most nuclear or cytoplasmic proteins.

flashBAC™ ULTRA

The most advanced flashBAC™ vector system, optimized by deletions in the chitinase, cathepsin, p10, p74 and p26 genes. The ideal system for the most difficult to express proteins including nuclear, cytoplasmic, membrane and secreted proteins.

flashBAC™ System으로 재조합 baculovirus를 제작하기 위해서는 insect cell, transfer plasmid, transfection reagent가 필요하다.
Mirus의 transfer vector (pOET1, pOET1C-6xHis, pOET6 BacMam) 외에도 다양한 transfer vector와 함께 사용할 수 있고 (호환 가능한 transfer vector list 보기), TransIT®-Insect Transfection Reagent (Code MIR 6104)로 높은 단백질 발현을 유도할 수 있다.
